Is the brass knuckle handle of this knife just a chuuni thing, or could it prove useful in a real fight? I know that brass knuckles can kill, I just don't see the point of trying to punch someone when you are holding a knife.

High doubt of it ever being useful if you're getting the knife in the position to punch someone with it. You're in position to stab someone and if someone is in basically hugging reach and is basically aware of you they'll likely try to twist your wrist and or force the knife out of your hand which case your other hand should be the one that's really punching.

Holy shit, guys. It's like a sharpened muzzle break. It's anything and everything that can cause damage when you HAVE TO cause damage. 

The knuckle hoops aren't just for punching, but to keep the weapon in hand. As last ng as you have a weapon in hand, your enemy has to adjust what he's doing to respond to that, limiting his options. If you're [moe]hugging,[/moe] you might not have the angle to effectively cut or stab with the blade, but if you can get it closer to him, the spikes can still grind against him, drawing blood, possibly severing a small artery or tendon. OR catching on webbing ot uniform and now you can control his body placement. 

And all that assumes he didn't do the smart/foolish thing if grabbing the knife wrist, allowing you to twist around and nick him in his wrist, which makes his fight a ticking clock against passing out.

AND AND all of your buddies with bayonets affixed behind you very probably seeing you locked up with him and looking to run him through long before either of you would even need a second wind.
